A role for natural killer cells in intestinal in£ammation caused by infectionwithSalmonella enterica serovarTyphimurium

چکیده
Acute gastroenteritis caused by Salmonella infection is a significant public health problem. Using a mouse model of this condition, the authors demonstrated previously that the cytokine gamma interferon (IFN-g) is required for a normal intestinal inflammatory response to the pathogen. In the present study, these experiments are extended to show that natural killer (NK) cells constitute an early source of intestinal IFN-g during Salmonella infection, and that these cells have a significant impact on intestinal inflammation. It was found that infection of mice with Salmonella increased both intestinal IFN-g production and the numbers of NK cells in the intestine and mesenteric lymph nodes. NK cells, along with other types of lymphocytes, produced IFN-g in response to the bacteria in vitro, while antibody-mediated depletion of NK cells in vivo resulted in a significant reduction in Salmonella-induced intestinal IFN-g expression. In a mouse strain lacking NK cells and T and B lymphocytes, intestinal production of IFN-g and Salmonellainduced intestinal inflammation were both significantly decreased compared with a strain deficient only in T and B cells. The authors’ observations point to an important function for NK cells and NK-derived IFN-g in regulating the intestinal inflammatory response to Salmonella.

Background and purpose: The aim of this study was to compare the proportion of natural killer (NK) and natural killer T (NKT)-like cells in dyspeptic disorders caused by Helicobacter pylori (H. pylori). Materials and methods: In a case-control study, 27 patients with gastric cancer (GC), 25 patients with peptic ulcer (PUD), and 22 patients with non-ulcer dyspepsia (NUD) were enrolled. After en...

Background: Pregnancy is a phenomenon that antigens of semi allogenic fetus are in direct contact with mother's immune system. Immune dysregulation can cause fetus rejection by mother's immune system responses. Human leukocyte antigen-G1, as an immunotolerant molecule has a major role to induce tolerance during pregnancy by suppression of natural killer cells through inhibitor receptors on thes...
متن کاملActivating natural killer (NK) cytotoxicity of canine CD5−CD21− cells requires low surface CD5 density NK cells
Natural killer (NK) cells play a crucial role in regulating immune functions. Few studies have characterized canine NK cells. We previously demonstrated that canine peripheral blood lymphocytes (PBLs) with a low surface CD5 density (CD5lo) are considered a critical NK population. Natural killer cells in most mammals do not express T-cell markers, but canine CD5lo cells express surface molecules...
متن کاملThe Efficacy of Toll-Like Receptors in Awakening Dendritic Cell/Natural Killer Cell System for Eradication of Tumors
Natural killer (NK) cells are effector cells of the innate immune system that exert direct cytotoxic functions. Ubiquitously-expressed toll-like receptors (TLRs) have been recognized as one of the major components promoting dendritic cell (DC) maturation, which may induce polarized immune responses beneficial to cancer immunotherapy. TLR-activated NK cells and DCs are prerequisite for robust ac...
متن کاملThe Effects of Dienogest on Macrophage and Natural Killer Cells in Adenomyosis: A Randomized Controlled Study
Objective Inflammation plays an important role in endometriosis and adenomyosis. Progestin has been used for symptomatic treatment of adenomyosis, although its effect on inflammation has not been studied. We conducted a randomized controlled study to investigate the changes of immune cell expression, i.e. macrophages and natural killer cells, in the tissue obtained from women with adenomyosis w...
